2006 Sportster 215hp scic

Only my #1 plug seems to be firing. Nothing happens or changes when #'s 2&3 are briefly disconnected. Could it be injectors? New fuel, new plugs, checked fuses. What next? Is it the ECU? Will pro equipment identify the problem or will they just change parts till it works?
